Aurigene's molecular biologists are well versed in cloning and expression
of cDNAs encoding chosen molecular targets. High level target protein
expression is achieved using bacterial, baculovirus / insect cells,
mammalian (transient and stable) or yeast systems. Expression of
target proteins in soluble and active form is accomplished by taking
advantage of a variety of novel vector and host strains.
Recombinant proteins are purified in large quantities from the suitable
expression systems utilizing traditional and affinity chromatography
methods. State-of-the-art protein purification instruments such
as FPLC are extensively utilized to purify proteins for screening
and structural biology studies.
Crystallization studies are performed for target proteins in complex
with known ligands as well as Aurigene synthesized / collaborator
compounds, supporting the SBDD group in the Lead Generation / Optimization
process.
